The above image is a snapshot of a how a protein is visualized in 3️⃣D space. The ‘ribbon’ model accurately portrays the way the amino acids are arranged in either an alpha-helix, a beta strand or more flexible or random loops. ⠀

I’ve been working on understanding the chemistry behind this specific protein for almost 5⃣ years. In this time I’ve been able to use techniques like NMR (Nuclear Magnetic Resonance) to demonstrate that certain amino acids (the coloured ⚪⚫🔴) are crucial for getting the protein to get it’s job done.⠀
⠀
Now, you may not light up with excitement over coloured ⚪⚫🔴like I do (or maybe you do!) but learning about how proteins do what they do is key to understanding how organisms work as a whole. ⠀
⠀
⌚ And we still have so much to learn!⠀
⠀
👖 We now know that the human genome contains approximately 20 000 genes that code for proteins – this is the proteome! Out of those 20000 about 17 – 18.5K have been identified. ⠀
⠀
Identified. Meaning that they have an ‘identity’ so we know they are there and the sequence but… how they look, work, and do what they do…? Let’s just say ‘PROCESS PENDING’.

Not only is the sheer number of proteins in the human proteome intimidating, so is the challenge of researching a protein in the lab. ⠀
⠀
Proteins are never static, they are very particular when it comes to choosing the type of environment they want to be in and who they want to hang out with in that environment. (Maybe I get those attributes from my proteins then ?🤔 )⠀
⠀
BUT the work in understanding these key players is important because knowing what a protein does and HOW it does what is does can be revolutionary in terms of our healthcare.⠀

Amazing things we take for granted everyday, like delivering oxygen throughout our hard-working muscles, breaking down our food into energy or even growing new cells, our 20000 proteins are working hard at making happen. When we know how proteins are working right we can understand whats happening when things go wrong.⠀

I sincerely hope that whatever he’s planning, I’m not involved in it... 🤦🏻‍♀️
On a more negative note, the 384 plate I ran the other day failed. Like, completely. Like evaporated samples.
I just want the weekend now, thanks. 👋🏼

At the start of the year I made a goal board with a mixture of work & life goals.📝

One of my crossfit goals is to be able to handstand walk. Due to my thesis being my main priority, 👩🏻‍💻I’ve seen crossfit more for fitness than trying to actively improve. It means I’ve not put enough work into achieving this skill anywhere near the amount I should have. So here I am trying to practice as much as I can before the year ends, just incase I can tick another one off soon! 💪🏼

Some road bumps have prevented me from achieving some of my goals this year, however, I have ticked 3/7 off and the others will be coming very soon, even if it will be in 2018. 👩🏻‍🎓🌏👣

I haven’t looked at my PhD in 9 months. Nine whole months! When I took time off with Nora (2nd maternity leave during PhD), I was determined it would be different – convinced I would find time to work. I even thought it could potentially buy me some time. How wrong was I? Nora is now 9 months old and I have done zero work. The PhD feels a like a distant memory! When I returned from maternity leave, there was so much else going on that I never got stuck back into the research. For one, there were numerous problems with the admin side of the PhD, and the many problems that come with taking maternity leave… twice (which seems to completely throw people)! I wasted so much time being passed from one person to the next – not the added stress that was needed in an already stressful time! Then, I got a full-time Lecturing position, and boy did the start of term hit me like a tidal wave (just as I was warned)! Again, I naively thought I would have time for the PhD, but that’s not happening. I am hoping Semester B will be more flexible (since I’ll have no module to lead), but that’s after the joy of marking 90+ assignments is out of the way! What the last 9 months has taught me, is that Stanley and Nora are my number one priority. I no longer have the same drive for the PhD, because everything goes into providing the best for these two. At times, I could easily give up on the PhD. I just don’t have the motivation. But then I tell myself I have come so far, and by reaching the end goal I could potentially provide more for them. I enjoy my job. I enjoy the research. I even enjoy teaching (more than I thought I would!). But, there are definitely times when I wonder what I am doing. I wish I could spend every minute with Stanley and Nora. I don’t want to miss anything. I want to enjoy this time whilst they are young... which is why more and more I am driven towards the idea of home-schooling. Even if we don’t achieve it within the next couple of years… it is most definitely something we are working towards 💛

I am looking for malaria parasite DNA from my bird blood samples, and the DNA has to be really good quality to be able to detect malaria during the next step... so, I am extracting DNA from the RNA samples we also got from each bird during the field season this year. .

After a meeting with my supervisor last week we agreed on some correlations I need to look at to strength my main results chapter. It means I have to use my mixed effects model code and tweak it to work for correlations. It’s been mind boggling.👩🏻‍💻

Yesterday afternoon/evening was super frustrating. Considering I’m on a tight thesis submission timeframe, having issues with statistics is far from ideal. 🙅🏻‍♀️ I didn’t crossfit and I worked until 9.30pm which I never do (I value my evenings). I still couldn’t get the damn code to work so to lower those stress levels prosecco was opened.👌🏽I promise I don’t normally resort to a glass of bubbles as a stress reliever! Mind you it was good.😏

Today has been a much better day. I woke up thinking if I stress it’s going to achieve nothing so I tested a few things and I spoke to another academic who is doing something similar and she helped to figure out the issue.💪🏼 Now to correlate, correlate and correlate some more data.

If something isn’t going right, take a break, deep breaths and come back to it with a fresh mind another day. And remember, seeking help is ok! ❤️

Agh Mondays.... It was negative degrees when I woke up and  really didn't want to get up before the sun today 🌄 #MondayBlues

By the time I'd got to the coffee shop I'd already reversed into the garbage bin and as soon as I arrived I realised I'd left my wallet at home and had no money. 
While I was trying to decide if I would drive home and come back or give up and go to the office, the owner let me know I could pay later. It was the sign I needed and I knuckled down and got my hour of research done. 
I am thankful for that peaceful time to work on my research because from 9:30 until 2:30 I didn't have a free moment. I finally grabbed lunch at quarter to 3.

In a year we've gone from fellow lab rats to fiancés to official Drs and now soon-to-be homeowners! It's all too grown-up, we might need a bouncy castle room or a jelly and ice-cream dispenser to balance things out a bit! ;) .

I find it so much easier to focus on small goals every day to reach the more significant, bigger goals.

I find that it makes the task less daunting, I achieve the end goal with less stress, I feel more productive and consequently motivated. 🙌🏼

Everyone has his/her own secrets to keep being productive. Mine is here: a good and diverse playlist of the favourite songs. After the lab move I was continuously struggling with the lack of motivation and productivity in the lab. Suddenly, my previous routines were not good anymore, and I had to deal with millions of small but annoying problems regarding stuff not working, reagents not arriving, impossibility to get things done in the time planned, etc. Probably, this was one of the most unproductive periods of my 2 year old Ph.D. So, I had to look for something to get back on a track, and music came extremely handy. It took away distructions, helped me to focus both during the labwork and data analysis, made boring stuff like preps, gels and hours of cell culture work, a bit more tolerable and, the most important, allowed me to control mood and stay positive. So, this is my advice: spend money on a good pair of the wireless headphones. It does pay off!

Remember the talk I was stressing about a few days ago? Well, turns out I was the guest of honour and speaker. 😊 I was invited by Nelson Mandela University to talk about my journey in science, importance of science communication, and changing the perceptions of academia. Then I got the opportunity to act as a judge during their Three Minute Thesis competition; and had the honour and privilege, together with the Deputy Vice Chancellor,  to hand out prizes to the 32 contestants and category winners. Phew, what an amazing experience!!! The passion the students have about their research is mind-blowing, I was so inspired!! I hope to continue with this wonderful relationship I have with Nelson Mandela University in the future. 😁

I would like to take this opportunity to thank the DST-NRF CoE in Palaeosciences for their support; my supervisor Dr Rose Prevec and my family for always encouraging me throughout this period. Recipe/Steps:
- boil water in a kettle or pot
- cut a small, thin slice of ginger and put it in a cup (I suggest a very small piece in the beginning and increase the quantity over time if you can handle it)
- pour boiling water over the ginger slice in the cup
- squeeze half a lemon in the cup
- wait 10-15 mins for the ginger to release it's medicinal properties into the hot water

♥ “When I got the chance to do research in my undergrad, I really LOVED it! I did a 2nd year, 3rd year AND a 4th year undergraduate thesis, so the natural path seemed for me, to pursue a PhD. I just wanted to continue doing what I loved to do and figured the career would work out for me afterward.”⠀

⚠ But this is not something Krishana necessarily recommends as the anxiety of post-graduation can be overwhelming.⠀

“In my 3rd year of my PhD I started to get anxious because I had no idea where my PhD would take me. I started to wonder ‘am I wasting my time?’ which eventually led to a very low mental state for me.”⠀

✨To overcome the anxiety of this uncertainty Krishana did what she loved to do – she researched and learned and this time not just from book and lectures but from people. ⠀

🎇“I wanted to educate myself on what else was out there so I took advantage of an internship at a pharmaceutical company where I met people in various fields like medical affairs, marketing, sales, regulatory affairs and this helped me a lot in changing my perception. I turned my ‘oh sh!t’ moment into something positive. This gave me confidence in myself, to see that the number of papers I published was not a measure of my success as a person”⠀

💪 Now Krishana is passionate about sharing that advice with you so that your hurdles can be easily jumped over. Her advice if you want to pursue graduate school? " Know your self-worth and don't compare yourself to other people"⠀
